GHK-Cu (glycyl-L-histidyl-L-lysine:copper(II)) is the naturally occurring copper complex of the GHK tripeptide, first isolated from human plasma. The Cu(II) ion is coordinated by the nitrogen from the imidazole side chain of histidine, the alpha-amino group of glycine, and the deprotonated amide nitrogen of the glycine-histidine peptide bond.

Research Dosage Notes

Research protocols typically use 1-2 mg/day via subcutaneous injection, 5 days/week, in cycles of 8-12 weeks followed by 2-4 weeks off. Some protocols use 100-200 mcg/day. Topical application is also common in cosmetic formulations. Reconstitution: 50 mg vial with 3 mL sterile water yields 16.67 mg/mL. Active at very low nanomolar concentrations. Long history of safe use in wound healing and skin care; naturally occurring and nontoxic.

Mechanisms of Action

5

Copper delivery and redox silencing

Strong

Copper(II) redox activity is silenced when complexed with GHK, allowing safe delivery of non-toxic copper into cells. Copper is required for over a dozen vital enzymes including those for connective tissue formation, antioxidant defense, and cellular respiration.

Copper transport / metalloenzyme activation

Collagen and ECM remodeling

Strong

Stimulates both synthesis and breakdown of collagen and glycosaminoglycans, and modulates the activity of metalloproteinases and their inhibitors, enabling dynamic tissue remodeling.

MMP / TIMP / ECM remodeling

Gene expression modulation

Moderate

Influences transcriptional networks associated with cellular migration, extracellular matrix turnover, neuronal survival, and redox balance. Modulates expression of thousands of human genes.

Multiple gene regulatory networks

Anti-inflammatory signaling

Moderate

Reduces levels of pro-inflammatory cytokines such as IL-6 and TNF-alpha while suppressing NF-kB-driven inflammatory cascades.

NF-kB / IL-6 / TNF-alpha

Angiogenesis promotion

Moderate

Stimulates blood vessel and nerve outgrowth, supporting tissue vascularization and innervation during repair processes.

VEGF / angiogenic signaling
