Research Dosage Notes

Subcutaneous: 200-500 mcg/day, titrated based on response. Topical: 0.005-0.1% concentration cream applied twice daily. Oral capsule and suppository forms are being explored for IBD. Generally well-tolerated with minimal side effects.

Mechanisms of Action

4

NF-kB inhibition

Strong

Blocks NF-kB activation at nanomolar concentrations, suppressing the master regulator of inflammatory gene transcription and reducing the cascade of pro-inflammatory cytokines (TNF-alpha, IL-1beta, IL-6).

NF-kB signaling

Melanocortin receptor activation

Moderate

Acts primarily through MC1R and MC3R receptors expressed on immune cells and epithelial tissues, triggering anti-inflammatory signaling without melanogenic (skin darkening) effects.

MC1R/MC3R signaling

PepT1-mediated intestinal uptake

Strong

Transported into intestinal epithelial cells and immune cells via the hPepT1 transporter, enabling targeted anti-inflammatory action in the gut mucosa.

hPepT1/intestinal epithelium

MAP kinase inhibition

Moderate

Inhibits MAP kinase inflammatory signaling pathways, further reducing pro-inflammatory cytokine secretion.

80

Gut inflammation reduction

Immune

Oral KPV reduces incidence of DSS- and TNBS-induced colitis in animal models, decreasing pro-inflammatory cytokine expression. Shows promise for IBD, ulcerative colitis, and Crohn's disease.

Strong
70

Epithelial barrier protection

Immune

Reinforces intestinal epithelial barrier integrity, preventing leaky gut scenarios where toxins and microbes cross into systemic circulation.

Moderate

Skin inflammation reduction

Immune

Topical application shows promise for psoriasis and eczema, reducing inflammation and hyperproliferation of keratinocytes.

Moderate
60

Systemic anti-inflammatory effects

Immune

Broadly reduces inflammatory markers across multiple tissue types, relevant to rheumatoid arthritis and other chronic inflammatory conditions.
