Pinealon is a synthetic tripeptide (Glu-Asp-Arg) developed by the St. Petersburg Institute of Bioregulation and Gerontology in Russia. It is based on the concept of bioregulatory peptides derived from pineal gland extracts, designed to influence gene expression in neuronal tissue.

Research Dosage Notes

Research dosages: in vitro studies used 10-100 nM concentrations; in vivo animal studies used 50-200 ng/kg injected over 5 days, or 10 mcg/kg daily for 5 days. Clinical studies used oral administration as an adjunct to standard therapy. No standardized human dosing protocol established. Commonly available as a lyophilized powder for reconstitution.

Mechanisms of Action

3

Direct DNA interaction and gene expression modulation

Emerging

Due to its ultra-small molecular size, Pinealon penetrates cell and nuclear membranes to interact directly with DNA, modulating chromatin structure and influencing gene expression related to neuroprotection.

Chromatin remodeling / gene transcription

ROS suppression and ERK 1/2 inhibition

Moderate

Suppresses reactive oxygen species (ROS) accumulation and inhibits ERK 1/2 activation, providing protective effects against oxidative stress in neuronal cells.

ROS / MAPK-ERK signaling

Neuroprotective signaling

Emerging

Exerts neuroprotective effects in cell culture and animal models, with demonstrated ability to maintain learning retention in diabetic rats and improve cognitive function in elderly patients and those with traumatic brain injury.

Cognitive enhancement and memory support

Cognitive

Clinical studies in elderly patients showed improved memory, enhanced performance efficacy, and correction of cerebral dysfunctions. Animal studies demonstrate maintenance of learning retention under diabetic conditions.

Emerging
60

Neuroprotection against oxidative stress

Cognitive

Prevents ROS accumulation in neuronal cells in a dose-dependent manner at concentrations of 10-100 nM, protecting against oxidative damage.

Moderate
50

Traumatic brain injury recovery support

Recovery

Clinical studies on patients with TBI consequences showed reduced duration and intensity of headaches, emotional balance improvement, and enhanced cognitive performance when Pinealon was added to standard therapy.

Emerging

Sleep optimization

Sleep

Reported benefits in sleep quality, likely related to its origin from pineal gland peptide research and its influence on neuronal signaling pathways.
