DSIP (Delta-Sleep-Inducing Peptide) is an amphiphilic nonapeptide first identified in 1977 from cerebral venous blood of rabbits during induced sleep. Note: this is the same peptide as 'Delta-sleep-inducing peptide' (id: 35) in this dataset.

Research Dosage Notes

Subcutaneous injection: typically 100-300 mcg administered in the evening before sleep. Clinical research used slow IV infusions at 25 nmol/kg. Very short half-life in vitro (~15 minutes) due to aminopeptidase degradation. Can cross the blood-brain barrier and is absorbed from the gut without denaturation.

Mechanisms of Action

5

Sleep architecture modulation

Moderate

Promotes delta (slow-wave) sleep patterns, though the exact receptor-level mechanism remains unclear. May selectively bind to regions of GABA-A, glycine, and/or NMDA receptors.

GABAergic / glutamatergic signaling

Stress hormone modulation

Moderate

Modulates cortisol and ACTH levels, potentially acting as a stress-protective peptide that normalizes the HPA axis.

Hypothalamic-pituitary-adrenal (HPA) axis

MAPK pathway interaction

Emerging

Interacts with components of the MAPK cascade and is homologous to glucocorticoid-induced leucine zipper (GILZ), affecting cell proliferation, differentiation, and apoptosis.

MAPK/ERK signaling

Pituitary hormone modulation

Moderate

Influences release of LH and GH from the pituitary, suggesting neuroendocrine regulatory roles.

Pituitary hormone secretion

Neurotransmitter modulation

Emerging

Influences levels and activity of serotonin (5-HT), glutamate, dopamine, and melatonin in the brain.

Monoaminergic / glutamatergic systems

Benefits

Improved sleep quality

Sleep

Promotes delta-wave (deep) sleep and may help normalize disturbed sleep patterns in insomniacs. Double-blind studies showed effects on chronic insomnia.

Stress reduction

Hormonal

Normalizes cortisol and stress response, potentially reducing anxiety and stress-related sleep disruption.

Pain modulation

Recovery

Some research suggests analgesic properties, possibly through opioid receptor interaction.

Hormonal regulation

Hormonal

May help normalize LH and GH secretion patterns, with potential downstream metabolic and recovery benefits.
